The very first step involved in this process includes the quantification of the medicinal substance with the help of a spectrophotometer. It is done in a transparent solvent and the absorbance is measured at a given wavelength. The wavelength which is generally selected is the wavelength belonging to the maximum absorption (λmax). In this step, a small error when encountered in the settling of the wavelength would have a minimum impact on the measured value of absorbance. In the next step, the concentration should be adjusted in such a manner that there is an absorbance of around 0.9. In this step, the calculation of the assay of a single component is carried out which consists of many other kinds of absorbing substances (Behera et al., 2012). This calculation can be done either with the use of standard absorptivity value, calibration graph or the single or the double point standardization. In the standard value absorptive method, standard A or E Values are used for doing the determination of the absorptiveness. This method proves to be useful when it is difficult to obtain any specific sample of the substance.
In calibration graph method, there is a measurement of the standard solution of the reference substance which are present at a concentration that would encompass he sample. After this, the construction of the calibration graph is done. The concentration is read with the help of the calibration graph itself. In single point standardization procedure, there is a measurement of absorbance of the sample solution and the measurement of another standard solution which is made of the reference substance.The concentration of the substances is calculation with the help of the proportional relationship between absorbance and the concentration. The material to be used for the purpose of research is Paracetamol. It is a drug and an inactive ingredient which is largely used in drug matrix. In order to do the experiment related to the analysis of paracetamol, diluents of methanol and water is used. Around 10 mg of drug is dissolved in 15 ml quantity of methanol l.85 ml of water is added to make the complete solution of 100 ml. From this solution, around 5 ml is taken and the volume is adjusted to around 50 ml with diluents.
